Note on working range: Values ka and kd are also dependent of each other, as well as the Bmax (binding maximum) values of the assay at the time, so in principle it is completely sample dependent range and the given ranges are just a guideline.
Extending working range: The range can be extended with careful experimental design. Please refer to J. Mol. Recognit. 2005; 18: 307–317 for more information.
Another option to extend the working range is Selectively Amplified Surface Plasmon Resonance (SAMP-SPR) technology that can be applied with SPR Navi™ instruments having additional wavelength (785 nm). SAMP-SPR is ideally suited to measure low concentrations of analyte in complex biological mixtures, with 100 times increased sensitivity and significantly reduced interference from background non-specific binding. This technology can be used to extend the range of affinity.
